For those of you who may not be interested in the science; this is a brief explanation to how Betaine (TMG) works in promoting an increase in lean mass.
1) Methionine loses methyl group and Homocysteine is created.
2) Increased homocysteine levels impair insulin signalling and thus the mTOR pathway (which increases protein synthesis) is impaired.
3) Glut4 may also be impaired due to reduced insulin signalling thus limiting glucose transportation into a muscle cell.
4) Homocysteine can be recycled back into methionine if it gains a methyl group.
Trimethylgycine is Glycine with 3 methyl groups attached. It donates a methyl group to homocysteine which is then recycled to make methionine.
5) Increase methionine levels also shown to increase nitrogen retention in pigs and thus signify an increase in lean muscle.
Betaine (TMG) also acts as an osmolyte: it maintains cell volume in a similar way to Creatine.
To understand the science behind Betaine Anhydrous, it is first important to understand Homocysteine.
What is Homocysteine and what does it do?
Homocysteine (HCY) is an amino acid which is produced from methionine once methionine loses its methyl group [3]. Elevated levels of HCY has been shown to impair insulin signalling [4], which can in turn reduce Akt (Protein kinase B) phosphorylation and thus impact protein synthesis [5].
Importance of the Akt (Protein Kinase B pathway (PKB))
PKB is able to induce protein synthesis via phosphatidylinositol 3-kinase (PI3K). PI3K activates PKB which then activates mTOR [6, 7, 8]. This process is facilitated through a pathway known as PI3K/ AKT/mTOR [6, 8]. mTOR is responsible for protein synthesis in humans and directly inhibits activation of cAMP and catabolism [6].
Furthermore, as insulin signalling is impaired by elevated HCY levels, Glut4 (an insulin regulated glucose transporter) will not be able to transport glucose into muscle cells, especially post exercise [9, 10].
The importance of Glut4 is recognised even more when examining diabetes patients who have developed a resistance to insulin [9, 10]. When the body is unable to recognise insulin, Glut4 is not activated and there is no longer an uptake of glucose into either muscle or fat cells [9, 10, 11]
It is because of this reason that hyperhomocysteinemia (elevated levels of HCY) has been linked to insulin resistance [9]. Where does Betaine (Trimethylglycine) fit into all of this?
Trimethylglycine (TMG) is the amino acid Glycine with three methyl groups attached to it and it is through these methyl groups where it serves a key function [12]. Methionine can create Homocysteine by losing its methyl group, however Homocysteine can also be converted back to Methionine upon receiving a methyl group [12]. TMG acts as this methyl donor and is thus able to reduce Homocysteine levels through this process [13, 14] TMG is also able to increase levels of S-Adenosyl Methionine (a universal methyl donating compound) which can then travel the body and donate methyl groups along In pigs, increased levels of Methionine correlated to an increase in Nitrogen rentention. This indicates a more positive nitrogen balance, lending to enhanced muscle growth [15].
Betaine (TMG) is also an osmolyte compound, meaning it plays a role in maintaining cell volume and fluid balance in a similar manner to Creatine [12, 14]
Through reduction of Homocysteine levels, insulin signalling is improved and thus the anabolic pathways are more readily activated. The end result of this process is improved body composition.

Suggested Use:
As a dietary supplement, tae 2.5 grams (2 and 1/2 scoops) 45 minutes to an hour prior to workout out or take 1.5 grams (1.5 scoops) prior to working out and 1 scoop spaced approximately 8 hours before or after. On non-workout days, take 1.5 grams (1 and 1/2 scoops) in the morning and 1 scoop in the evening.
Betaine Anhydrous can be taken with or without food.
